Effects of Wumei Wan on interleukin-lp, phosphorylase nuclear factor - κB and forkhead transcription factor 3 in rats with immune colitis.

- Abstract
Objective To observe the influence of Wumei Wan (MENume Pill) on the expressions of interleukin-11 (IL-1β), phosphorylase nuclear factor-κB (pNF-κB) and forkhead transcription factor 3 (Foxp3) in serum and colon tissue in rats with immune colitis, and discuss the effective mechanism. Methods SD rats (n = 40) were randomly divided into normal group, model group, methylprednisolone group (MPSL group, 2.0 mg/kg), low-dose Wumei Wan group (13.4 g/kg) and high-dose Wumei Wan group (26. 8 g/kg, each n = 8). The model of immune colitis was established through intracolonic instiilation of 2,4,6-trinitrobenzenesulfonic acid (TNBS) and ethyl alcohol. The rats in all groups were orally given corresponding medicinals once a day for 14 d after modeling for 3 d and then all indexes were detected. The colon mucosa damage index (CMDI) was observed after anatomy. The level of IL-1β was detected by using ELISA, protein expression of pNF-κB was detected by using Western blotting assay, and gene expression of Foxp3 was detected by using qPCR in all groups. Results Compared with normal group, the level of serum and colon IL-1β increased, and protein expression of colon pNF-κB increased and expression of colon Foxp3 decreased in model group (P < 0. 05). Compared with model group, level of serum and colon IL-1β increased and expression of Foxp3 increased in MPSL group (P <0. 05) . The level of colon IL-1β and protein expression of pNF-κB decreased, and level of serum IL-1β and expression of colon Foxp3 increased in low-dose Wumei Wan group and high-dose Wumei Wan group (P < 0.05). Compared with MPSL group, the level of serum IL-1β increased (P <0. 05) and level of colon IL-1β decreased (P < 0. 05) low-dose Wumei Wan group and high-dose Wumei Wan group. Conclusion Wumei Wan can down-regulate the expressions of pNF-κB and IL-1β in damaged colon tissue through increasing Foxp3 expression, and thereby relieve colon pathological damage and prevent the progression of immune colitis.
