- Abstract
F430, ein einzigartiger enzymatischer Kofaktor, ist an der Bildung und Oxidation von Methan in streng anaeroben Bakterien beteiligt. Das Schlüsselenzym Methyl‐Koenzym‐M‐Reduktase (MCR) weist in seinem aktiven Zentrum einen porphinoiden Nickel‐Komplex auf, der ein charakteristisches Absorptionsmaximum bei etwa 430 nm zeigt. Hier berichten wir über die dreistufige, von Vitamin B12 ausgehende Partialsynthese eines hybriden NiII‐haltigen Corrins, dessen strukturelle und spektroskopische Eigenschaften teilweise denen von F430 ähneln. Schlüsselschritt der Syntheseroute ist die simultane Demetallierung und Ringschlussreaktion eines 5,6‐Secocobalamins zum metallfreien 5,6‐Dihydroxy‐5,6‐dihydrohydrogenobalamin mittels Cobaltocen und KCN unter reduktiven Bedingungen. Untersuchungen zur Koordinationschemie der neuartigen Verbindung untermauern die frühere Hypothese, warum die natürliche Selektion des Corphinliganden gegenüber dem Corrinliganden in F430 es ermöglichte, anspruchsvolle Ni‐katalysierte Reaktionen in biologischen Systemen durchzuführen. An der Bildung und Oxidation von Methan in streng anaeroben Bakterien ist der Kofaktor F430 beteiligt. Das Schlüsselenzym Methyl‐Koenzym‐M‐Reduktase enthält in seinem aktiven Zentrum einen porphinoiden Nickel‐Komplex. Eine von Vitamin B12 ausgehende, dreistufige Partialsynthese liefert ein hybrides, NiII‐haltiges Corrin mit strukturellen und spektroskopischen Eigenschaften ähnlich denen von F430.
