- Abstract
Milk contains many bioactive compounds, in which exosomes have been paid more attention recently because of its potential to regulate physiological functions by transferring encapsulated cargos to target cells. The bovine milk exosomes could be absorbed through passing the intestinal barrier and further reach the systemic circulation. In this study, we isolated exosomes from bovine milk by centrifuge and incubating with ExoQuick-TC reagent to evaluate the optimal isolating conditions. The isolated exosomes were analyzed by Bradford protein assay, scanning electron microscope, nanoparticle tracking analysis, and immunoblot. The results revealed that most of milk proteins were removed and incubating with ExoQuick-TC for 8 h had the most exosome protein concentration. In conclusion, using centrifuge with ExoQuick-TC can obtain more exosomes with marker protein. Thus, the method will be helpful for isolation of exosomes from bovine milk and contribute to the progression of further studies.
