- Abstract
Introdução:Os eritrócitos são muito suscetíveis ao estresse oxidativo, uma vez que suas membranas são ricas em ácidos graxos poli-insaturados; estas células têm altas concentrações de hemoglobina e oxigênio, que podem agir como promotores do processo oxidativo (auto-oxidação); a citólise eritrocitária per si já fornece um sinal analítico com a liberação da hemoglobina. Em condições de estresse oxidativo, os eritrócitos estão entre as primeiras células do organismo a serem afetadas, este processo pode causar danos na membrana celular eritrocitária podendo levar a disfunção do eritrócito.Desta forma, os eritrócitos têm sido utilizados como sistemas celulares para estudo dos mecanismos de oxidação gerados por espécies reativas e consequentemente para explorar o potencial oxidante ou antioxidante de compostos naturais ou sintéticos, sistemas enzimáticos e/ou procedimentos terapêuticos e analíticos.Para monitorar a lipoperoxidação (LPO) em biomoléculas, a abordagem mais utilizada é a quantificação de substâncias reativas ao ácido tiobarbitúrico (TBARS), pois é relativamente simples, rápida e de baixo custo. Entretanto, a avaliação de TBARS em eritrócitos é complexa e difícil pois há possíveis interferências da hemoglobina e da presença de ferro que pode catalisar reações de oxidaçãona amostra (Kanias e Acker, FEBS J., v. 277, p. 343-56, 2010). O objetivo deste trabalho foi comparar a eficácia entre o AAPH e o t-BHPcomo indutores de LPO utilizando eritrócitos humanos em modelo experimental.Metodologia:Utilizou-se amostras de sangue venoso com EDTA de indivíduos saudáveis(Certificado de Apresentação de Aprovação Ética: n° 76883417.0.0000.5426)eapós lavagens com PBS, preparou-se um concentrado de hemácias ("papa" de hemácias) o qual foi utilizado nos ensaios para investigaros níveis de TBARS, na ausência ou presença de AAPH (50mM) que gera radicais por termólise ou t-BHP(0,4mM). Resultados e discussão: Nos ensaios realizados na presença AAPH obteve-se baixo sinal analítico (absorbância ou fluorescência), com valores próximos a ausência do AAPH (background).Todavia, nos ensaios na presença dot-BHP observou-se um maior sinal analítico tanto para absorbância quanto para fluorescência, quando comparado ao seu background(ausência de t-BHP). Conclusão: De acordo com os resultados, observou-se que a metodologia utilizando o t-BHP como indutor da LPO nos eritrócitos tem sinal analítico mensurável, por espectrofotometria de absorção ou de fluorescência, o suficiente para as análises fisiopatológicas ou como sistema exvivode agentes oxidantes ou antioxidantes.
