- Abstract
Extraction and purification techniques of total saponins from bulbs of Lilium brownii F. E. Brown var. viridulum Baker. and the antidepressant activity of the total saponins were studied in this paper. Based on orthogonal design, four factors in extraction of saponins from bulbs of Lilium brownie were optimized : concentration of ethanol, reflux time, solid/liquid ratio and extraction times. The optimized process for extracting total saponins from bulbs of Lilium brownii was confirmed as follows : reflux time, 3 h; alcohol, 80% ; solid/liquid, 1 : 10 ; extraction 3 times. The highest extraction rate reaches 1.24%. Then, the extract of total saponins was separated and purified by AB-8 porous resin absorption and the total saponins content reaching 62%. In antidpression effect filtration study, the mouse tail suspended experiment, mouse forced swimming experiment were performed, the result showed that the total saponins was the main antidepression component. The above results showed that the total saponins had certain antidepressant activity.
