- Abstract
In order to e explore the infection outcomes and pathogen nucleic acid loads in different breeds of sheep infected with Mycoplasma pneumoniae (Mo), 5 sheep breeds were infected with Mo Guizhou Weining (GZ-WN) strain. The pathological changes of each tissue were observed after infection or sacrificing, and the nucleic acid load of Mo in each tissue was detected by fluorescence quantitative PCR. The nucleic acid load of Mo in different tissues of the same breed of sheep and the nucleic acid load of the same tissue of different breeds of sheep were statistically analyzed. The results showed that Mo could cause infection in five breeds of sheep, and the mortality rate ranks as follows: white goat≈black goat>Ma sheep>Boer goat>Hu sheep. Mo infection could cause pathological changes of different degrees in lung, trachea and thorax of five breeds of sheep; lung adhesion and tracheal mucus lesion of black goat was more serious than that of other breeds; pleural effusion of white goat was more serious than that of other breeds, followed by black sheep. The heart, liver, spleen and kidney of the five breeds of sheep were not visible to the naked eye. The results of statistical analysis of Mo nucleic acid load in different tissues of the same sheep breed showed that the Mo nucleic acid load was different in different tissues of the same breed, and the Mo nucleic acid load in the trachea of each breed was the highest, which was significantly higher than that in other tissues (p<0.01), followed by lung, while Mo load in spleen and kidney was lower. The statistical results of Mo nucleic acid load in the same tissue of different breeds of sheep showed that the Mo nucleic acid load in the trachea of the five breeds was the highest in Boer goat and the least in white goat, which was significantly lower than that in other breeds (p<0.01); the Mo nucleic acid load in lung of black goat was significantly higher than that of other breeds (p<0.05); and the Mo load of Hu sheep was the lowest. In heart tissue, the nucleic acid load of Mo in black goat was significantly higher than that in other breeds of sheep except Ma sheep (p<0.05), and the lowest was white goat. In liver tissue, the nucleic acid load of Mo in Ma sheep was the highest, which was significantly higher than that in other breeds except Hu sheep (p<0.05), while that in white goat was the least. In spleen, the nucleic acid load of Mo was the highest in Hu sheep, significantly higher than that in other breeds of sheep (p<0.05), and the least in Ma sheep. In kidney tissue, the Mo nucleic acid load of Hu sheep was the highest, which was significantly higher than that of other breeds of sheep except Boer goat (p<0.05), and the least was in Ma sheep. The above results showed that there were differences in tissue pathological changes and tissue nucleic acid load of Mo infected sheep of the same breed, and the nucleic acid load of Mo in infected sheep organs was positively correlated with the degree of tissue pathological changes and clinical manifestations. The Mo nucleic acid load of the same tissue was different in different breeds of sheep infected with Mo. To date, there has been no domestic or foreign studies on the difference of histopathology and nucleic acid load of artificial infection with Mo in different breeds of sheep. The results of this study provide a reference for exploring the mechanism of different infection outcomes by Mo in different breeds of sheep.
