- Abstract
Objective: To investigate the effect and mechanism of curcumin on elevating of oxidative stress and endothelial dysfunction induced by high glucose. Methods: The impaired Human Umbilical Vein Endothelial Cells (HUVECs)model was induced by cultured with high glucose DMEM (33m M D-glucose). The ROS level was detected by DHE fluorescent probe 24 h after treated with or without curcumin. Aorta from C57BL/6J mice were isolated and cultured with high glucose DMEM, low glucose DMEM, high glucose DMEM plus curcumin or high glucose plus curcumin and compound C respectively for 24 hrs. Wire myograph was used to analyze the vascular function. And phosphorylation of e NOS and AMPK was detected by western blotting. Results: Curcumin ameliorated the ROS level induced by high glucose, and Com C reversed this situation. Cultured with curcumin increased p-e NOS level and improved the endothelium dependent relaxation significantly but decreased the endothelium independent relaxation under high glucose condition, these results also blocked by cultured with Com C. Curcumin increased the p-AMPK level of high glucose treated ECs. Conclusion: Curcumin attenuated ROS level and improved endothelium dependent relaxation in high glucose treated ECs and aortic, these effects may be due to the activation of AMPK/e NOS pathway.
