ESTUDIO DE ASOCIACIÓN DE VARIANTE GENÉTICA DE BDNF Y LONGITUD TELOMÉRICA EN PACIENTES CON DEMENCIA ALZHEIMER Y CONTROLES.

- Abstract
Objetivo: Conocer si existe asociación entre la LT y la variante genética BDNF_rs6265 con la cognición y síntomas comportamentales en pacientes con DA. Antecedentes: La demencia tipo Alzheimer (DA) se caracteriza por alteraciones cognitivas y comportamentales. La DA se ha asociado con varios biomarcadores de deterioro/envejecimiento biológico como la longitud telomérica (LT), y los niveles del factor neurotrófico derivado del cerebro (BDNF) con la cognición y el comportamiento. Una disminución en la LT y la variante rs6265 del gen BDNF, también se han relacionado con deterioro cognitivo y comportamental en la DA. Sin embargo, estas asociaciones aún no están completamente comprendidas. Métodos: Se aplicó entrevista clínica, inventario neuropsiquiátrico de Cummings, prueba cognitiva (MoCA) y escala de trastorno de sueño para el adulto mayor a los participantes. A partir de DNA de sangre periférica se genotipó la variante de BDNF_rs6265 y se cuantificó la LT por QPCR. Las frecuencias alélicas-genotípicas de la variante se determinaron en 100 controles sanos (CT) adicionales. Se analizaron estadísticamente los datos clínicos-moleculares. In silico, se evaluaron las interacciones de BDNF con otros genes de riesgo (APOE y circadianos) previamente estudiados. Núm. de registro del protocolo: 11/20. Resultados: Se evaluaron 28 CT (edad 70+8 años) cognitivamente sanos y 32 pacientes con DA (edad 73+9 años). La LT (0.94+0.86) de CT fue mayor que en los pacientes con DA (0.91+0.64), p=0.05. Hubo una correlación negativa entre la LT y edad (r=-0.43; p=0.02) en CT. La LT se relacionó con conducta ansiosa (p=0.043) en los pacientes. Conclusiones: En los pacientes con DA, la LT presentó asociación con síntomas de ansiedad. Mientras que, la variante de BDNF_rs6265 no presentó asociación con las variantes analizadas en esta muestra, y tampoco se observó interacción in silico con otras variantes de riesgo para DA.
