- Abstract
Objetivo: determinar si la modulación de los niveles de ácido kinurénico (KYNA) por la N-acetilcisteína (NAC) previene el deterioro cognitivo inducido por la exposición postnatal temprana a plomo en roedores. Antecedentes: en roedores, la intoxicación por plomo (Pb2+) durante etapas tempranas del neurodesarrollo induce deterioro cognitivo e incrementa los niveles cerebrales de KYNA. Debido al antagonismo de KYNA sobre los receptores nicotínicos y NMDA, su incremento induce alteraciones cognitivas. Siendo que NAC inhibe a la kinurenina aminotransferasa II (enzima para la neosíntesis de KYNA), representa una herramienta para reducir el KYNA cerebral. Métodos: los tratamientos se realizaron durante la lactancia (día 0-23 posnatal (PN)) en los siguientes grupos: 1) control, 2) Pb2+, 3) NAC y 4) Pb2++NAC. El NAC (350 mg/día) fue administrado en el alimento a los grupos 3 y 4; adicionalmente los grupos 2 y 4 recibieron 500 ppm de acetato de plomo en agua y el grupo 1 recibió agua potable y comida regular. Las pruebas cognitivas de ubicación de alimento enterrado (UAE) y objeto novedoso se realizaron al día 60 PN y se evaluó la lipoperoxidación, los niveles de GSH y KYNA, además de la actividad de KAT II en el tejido cerebral.Núm. de registro del protocolo: 111/19. Resultados: los ratones intoxicados con Pb2+ no discriminaron entre el objeto novedoso y el conocido, exhibiendo alteraciones en la consolidación de la memoria en ambas pruebas realizadas. Estas alteraciones cognitivas fueron atenuadas por la coadministración con NAC; que además redujo las alteraciones redox y los niveles cerebrales elevados de KYNA inducidos por Pb2+. Se encontró una correlación positiva entre la distancia recorrida (en UAE) y los niveles cerebrales de KYNA. Conclusiones: estos datos demuestran el potencial de la modulación de KYNA para reducir las alteraciones cognitivas inducidas por la exposición postnatal temprana a plomo.
