Participación de la víia del ON-GMPc-PKG-canales de K<sup>+</sup> en el efecto antialodínico de resveratrol en un modelo experimental de dolor neuropático.

- Abstract
Introducción: las lesiones en el sistema nervioso central o periférico producen dolor neuropático, que se manifiesta por trauma (cirugía, amputación), infecciones (SIDA, herpes), toxinas (vincristina, cisplatino, talidomida, As, Pb, Au, Hg), trastornos metabólicos (diabetes mellitus, hipotiroidismo), lupus eritematoso, artritis reumatoide y epilepsia. Los síntomas característicos del dolor neuropático son: alodinia (dolor ante un estímulo que no debería ser doloroso, como el roce de la ropa) e hiperalgesia (dolor incrementado ante un estímulo doloroso). Los analgésicos no esteroídeos y opioides no alivian de manera efectiva este padecimiento, por lo que se utilizan fármacos alternativos para su tratamiento (gabapentina, baclofén, carbamazepina, valproato, amitriptilina). En la búsqueda de nuevos fármacos que alivien el dolor neuropático, en este trabajo se evaluó la posible participación de la vía del óxido nítrico (ON-GMPc-PKG-canales de K+) en el efecto antialodínico de resveratrol (Rv), y la modificación de la actividad de las sintasas de óxido nítrico (SON) en médula espinal. El Rv es una fitoalexina presente en uvas, se utiliza como suplemento alimenticio y presenta actividad antioxidante, anticancerígena y antihipertensiva. Estudios recientes indican que este compuesto produce un aumento en la expresión del RNAm de las SON, activa a la enzima guanilato ciclasa (GC) e induce un incremento en la corriente de canales de K+ activados por Ca2+ de alta conductancia (BKCa) en células endoteliales, ya que la apertura de estos canales en neuronas sensoriales induce alivio del dolor. Material y métodos: la Inducción de neuropatía se realizó en ratas Wistar hembra, a las cuales se les ligaron los nervios lumbares izquierdos 5 y 6. Al décimo día se les canuló intratecalmente, y cinco días después se aplicó la prueba de umbral táctil con filamentos "Von Frey". Se administró en la espina Rv (100-600 µg), la dosis más efectiva de Rv se coadministró con cada inhibidor o bloqueador: L-NAME (10-100 µg; inhibidor no selectivo de las SON), ODQ (1-10 µg; inhibidor de la GC), KT-5823 (5-500 ng; inhibidor de la proteína cinasa G, PKG), íberiotoxina (IbTx, 5-500 µg; bloqueador de canales BKCa), glibenclamida (Glib, 12.5-50 µg; bloqueador de canales de K+ sensibles a ATP, K+ATF). El efecto antialodínico se expresa como área bajo la curva (µ/min). La participación det ON en el dolor neuropático se analiza indirectamente mediante la técnica de Bredt y Snyder (1990), en donde se mídela actividad de la SON evaluando la conversión de [3H]- L-arginina a [3H]-L-citrulina. Ratas neuropáticas controles y tratadas con Rv a 1, 2, 2.5, 3 y 5 horas fueron sacrificadas por decapitación. La región lumbar de la médula espinal fue disecada en secciones dorsal y ventral, lados izquierdo y derecho. Las muestras de médula espinal se procesaron para evaluar la actividad de las SON, que se reportó como ng 3H-L-citrulina/ 500 µg de proteína/1 minuto. En el análisis estadístico se utilizó ANOVA de una vía y la prueba de Dunnet con p<0.05. Resultados: la administración espinal de 300 µg Rv produjo un efecto antialodínico más efectivo y este efecto se revirtió por L-NAME (100 µg), ODQ (10 µg), KT-5823 (500 ng) e IbTx (500 ng).…
