- Abstract
Treatment results of malignant tumors, including multiple myeloma show great divergence even within the same histological (sub)type. Basis of this phenomenon is the different genetic background of individual tumors. Recent scientific advance highlighted the fact that dysfunction of the tumor suppressor gene TP53 results in extensive therapy-resistance. In this review, we attempt to provide a cell biology oriented insight for clinician readers aimed at appreciating the complex biological role of protein p53, the consequences of its molecular and cytogenetic alterations; to better appreciate the received laboratory results. Afterwards, treatment results of myeloma patients with mutant or haploinsufficient TP53 are detailed, together with the critical analysis of presently available disappointing and possibly more effective future therapeutic options.
