- Abstract
The separation and purification technology of cAMP from Xinjiang Hami jujube was studied. By comparing the static adsorption and desorption of four macroporous resins,the best resin for purification of cAMP in the extract of Hami jujube was determined. By dynamic adsorption and elution of macroporous resin, the factors such as sample concentration,pH and flow rate were investigated to determine the best purification process of cAMP. The results showed that HZ-802 resin had better purification effect. The optimum conditions for dynamic adsorption were as follows:sample volume 220 mL,flow rate 2.0 mL/min,concentration 20 μg/mL and pH 5 of sample solution. The optimum conditions for dynamic analysis were as follows:40 % ethanol in eluent,120mL in eluent and 3mL/min in eluent. After purification,the concentration of cAMP was 38.24 μg/mL, and the content of cAMP in the crude extract was 0.10 % after freeze-drying.
