- Abstract
The article focuses on the study on the development of a rapid and high-throughput genotype method for the detection of benzimidazole-resistant allele E198A in Sclerotinia sclerotiorum population through the developed mismatch primers for Allele-specific nucleotide polymerase chain reaction with mismatch primers (ASQPCR-MP). It dwells on the analysis on the genomic DNA from 223 sclerotia. It reveals that ASQPCR-MP has the capacity to differentiate benzimidazole fungicidesHR and benzimidazole-sensitive MBCS phenotypes. It concludes that ASQPCR-MP is suitable for epidemiological studies related to high-throughput genotype formats. A comparison on the assays between mycelial growth inhibition, ASPCR, and ASPCR-MP is provided.
