- Abstract
Pain is one of the most commonly reported and least understood symptoms amongst cancer patients and survivors. Improper management of pain can lead to significant reductions in physical and psychological wellbeing, and diminished quality of life. Patients with similar types and stages of cancer can present vastly different accounts of severity of pain, causes of pain, and responses to treatment methods aimed to reduce pain such as radiation therapy and analgesics. These differences suggest an underlying genetic etiology, and highlight the lack of understanding of how cancer pain develops. The present selective review summarizes 25 studies on biomarkers of pain in cancer patients. Many of these studies show that up-regulation of inflammatory biomarkers and hormones is associated with increased reports of pain and pain severity. In addition, single nucleotide polymorphisms within common inflammatory genes and signaling protein genes have been associated with risk for cancer pain. The review concludes that there are a variety of biomarkers with clinical utility in predicting cancer pain, and may also act as potential therapeutic targets. Future studies should look to examine their role in the progression and development of pain, and should also look for biomarkers that can distinguish between the different types of cancer pain.
