- Abstract
Introdução: A obesidade é atualmente considerada uma epidemia global. O aumento nas taxas de obesidade e sobrepeso é considerado uma preocupação urgente de saúde pública, além de um fator de risco importante para o risco de síndrome metabólica. O excesso de lipídios favorece o desequilíbrio energético, levando a um aumento do estoque de gordura corporal e remodelamento do tecido adiposo, o que está diretamente associado à indução da obesidade. O tecido adiposo marrom (TAM) é um tecido metabolicamente ativo e está relacionado com a dissipação de energia na forma de calor durante a termogênese, contribuindo para o gasto energético. Na obesidade, o acúmulo lipídico no TAM é considerado a principal causa de disfunção desse tecido. O extrato hidroalcóolico do caroço do açaí (ASE) apresenta um importante conteúdo polifenólico, como catequinas, epicatequinas e proantocianidinas poliméricas, que estão relacionados aos seus efeitos antiobesidade, cujos mecanismos ainda não estão bem estabelecidos. Portanto, o objetivo deste trabalho é avaliar os efeitos do tratamento com ASE sobre o remodelamento e disfunção do tecido adiposo e o seu papel na indução da termogênese, e na biogênese mitocondrial no TAM. Metodologia: O uso de animais foi aprovado pelo comitê de ética da UERJ, sob o número 004/2021. Camundongos machos C57BL/6 foram separados em três grupos: controle (dieta 10% lipídeos), HF (dieta 60% lipídeos) e HF+ASE (dieta 60% lipídeos + 300 mg/kg/dia por gavagem intragástrica). A dieta foi administrada concomitantemente ao tratamento durante 12 semanas. A massa corporal foi avaliada ao início e ao final do período de tratamento. As alterações morfológicas do TAM foram analisadas histologicamente. Lâminas de TAM também foram utilizadas para imunohistoquímica, com marcação para UCP-1. Foram avaliadas a expressão de marcadores relacionados à indução da termogênese, ativação do TAM e biogênese mitocondrial em homogenato de TAM através de western blot. Resultados: O ASE preveniu o ganho de massa corporal e as alterações morfológicas no TAM dos animais tratados com o extrato quando comparados ao grupo HF. Em TAM, o tratamento com o ASE promoveu o aumento da expressão dos marcadores envolvidos na indução da termogênese (UCP-1 e ß3-AR). O aumento da expressão proteica de UCP-1 no grupo tratado com ASE foi corroborado pela imunomarcação desta proteína em lâminas de TAM. O ASE também promoveu aumento na expressão dos marcadores envolvidos na ativação do TAM (pAMPK, pLKB1 e SIRT-1) e biogênese mitocondrial (PGC-1a, NRF1 e CPT-1) do TAM em relação ao grupo HF. Conclusão: Esses achados demonstram que o tratamento com o ASE preveniu as alterações estruturais em TAM de camundongos submetidos à dieta hiperlipídica, além de aumentar a expressão de proteínas chaves relacionadas à indução da termogênese e à biogênese mitocondrial, sugerindo que o extrato pode ser utilizado como uma abordagem importante na prevenção da obesidade.
