- Abstract
Objective To observe the effect of St. John’s wort extract on a mouse model of chronic unpredictable stress-induced depression and examine its mechanism. Methods The chronic unpredictable stress method was adopted for four weeks to establish the mice model of depression. Test animals were randomly divided into normal control,model control,fluoxetine hydrochloride,and St. John’s wort extract low,medium and high( 0.2 g/kg,0.4 g/kg,0.8 g/kg,respectively) dose groups. The sugar water preference of the mice was determined. To induce stress,different conditions after the last dose of autonomous activities were employed were four weeks, and a tail suspension test was performed. Blood was taken to detect the serum 5-HT and NE levels,and the pathological changes in the hippocampus were observed. Results The St. John’s wort extract low,medium,and high dose groups could reverse the above changes( P < 0.05 or P < 0.01),had increased serum 5-HT and NE levels( P < 0.05),and reduced neuronal damage in the hippocampal tissue region. The effects observed in the fluoxetine hydrochloride group were similar. Conclusion St. John’s wort extract had a noticeable anti-inhibitory effect on the mouse model of depression caused by chronic unpredictable stress. Its effect may be related to the alleviation of neuronal damage in the hippocampal tissue by regulating the levels of the neurotransmitters,5-HT and NE.
