- Abstract
Amyotrophic Lateral Sclerosis (ALS) is a progressive and fatal neurodegenerative disease affecting motor neurons responsible for controlling voluntary muscles. Advances in research have led to a greater understanding of the underlying mechanisms, including genetic, cellular, and molecular aspects, that contribute to the development and progression of ALS (Al-Chalabi & Hardiman, 2013). The diagnosis of the disease remains a challenge, and emerging biomarkers, such as specific proteins in cerebrospinal fluid and plasma, and advanced imaging techniques, such as magnetic resonance imaging, are being investigated to improve diagnostic accuracy and disease progression monitoring (Turner et al., 2009); (Foerster et al., 2013). Furthermore, various experimental therapeutic approaches, such as gene therapy, stem cell therapy, immunotherapy, and RNA therapy, are under development and in different stages of clinical trials (Brown & Al-Chalabi, 2017); (Petrov et al., 2017). This article presents a detailed and in-depth review of the mechanisms, diagnosis, and potential treatments for ALS, highlighting recent advances, challenges, and future directions in research and treatment of this complex neurodegenerative disease.
