- Abstract
Immunofluorescence assay (IFA) The expression of US9 protein ininfected Sf9 cells was detected using theindirect immunofluorescence assay (IFA).The reaction showed specific reactivitywith the anti-BoHV-1 positive serum forvAc-US9-infected cell, while no reactivitycould be observed in non-infected cells(mock-infection). Indirect US9- based ELISA test The indirect US9-based ELISAwas performed in order to verify theantigenic properties of the US9 antigenexpressed by the recombinant virus in Sf9insect cells. Thecrude lysate from the infected Sf9 cellswith vAc-US9 as coating antigen provedto be sensitive and efficient in indirectELISA test for detection of anti-US9serum samples infected with BoHV-1.The reactivity of vAc-US9/BoHV-1 withanti BoHV-1 positive serum in indirectELISA indicated that, the baculovirusexpressed US9 was retained its antigenicproperties as well as the native US9 ofBoHV-1. The photo shows infectedSf9 cells using the recombinant virus expressing US9 protein (vAc-US9)which was reacted with anti BoHV-1 positive serum and probed with FITC-labelledanti-BoHV-1.
