Maternal Manganese Restriction Transiently Impairs Glucose Tolerance and Insulin Secretion in Male Rat Offspring.

- Abstract
Maternal mineral restriction is important in programming the body composition (adiposity) and insulin secretion in the rat offspring. We studied the effects of maternal restriction of Manganese an essential trace element that modulate gluconeogenesis, insulin biosynthesis and signaling, in impairing glucose tolerance and insulin secretion in offspring. Female, weanling, Wistar/NIN rats received for 3 months a control or Mn restricted (95 %) diet and mated with control males. Some restricted mothers were rehabilitated with control diet from conception (MnSC), parturition (MnSP) and their offspring raised on control diet. Some restricted offspring were weaned on control diet (MnSW) while others continued on restricted diet throughout (MnR). Oral glucose tolerance and plasma insulin levels were determined in offspring of both sexes at 3, 6 and 9 months of age. MnR per se did not affect fasting glucose in male offspring whereas MnSC, MnSP and MnSW induced significant and consistent fasting hyperglycemia. At six months of age MnR offspring showed fasting hypoinsulinemia and no rehabilitation regime could correct it. However, glucose AUC during OGTT was higher in MnR at 6 months; MnSC and MnSP but not MnSW could correct this effect. Also, MnR per se decreased Insulin AUC at 3 and 6 months of age, which was not mitigated by MnSC, MnSP and MnSW. There were no differences among offspring of different groups at 9 months in any parameter suggesting that Maternal Mn restriction may impair glucose tolerance and insulin secretion in offspring but the changes were transient and disappear without / with rehabilitation by 9 months. Similar transient changes were seen even in female MnR offspring at 3 months but were partially mitigated by the rehabilitation regimes. Again by 9 months, all the parameters were comparable among groups. Maternal and postnatal Mn restriction may thus impair transiently, glucose tolerance and insulin secretion in the offspring.
