Preparation and characterisation of phytosomes of cassia fistula extract.

- Abstract
The research work aimed to prepare phytosome- a combined drug delivery system and classify Cassia fistula extract phytosome vesicles. Analysis of X-ray diffraction (XRD) and Differential Scanning Colorimetry (DSC) was performed to study interaction between the various excipients used in phytosome preparation and Cassia fistula extract. Modified thin-film hydration technique prepared the phytosomal formulation. Phospholipids (soylecithin to cholesterol) were dissolved in 3ml of organic solvents (chloroform and methanol) in a ratio of 2:1 for organic phase. The solvent phase was evaporated by gentle 40 ° C heating. The resulting dried mixture was redispersed in 10 ml deionized water containing 50 mg of Cassia fistula extract. The resulting mixture was further sonicated using a 10-minute probe sonicator of 5 sec ON / OFF period and 30 ° C amplitude. The optimised formulation revealed internal vesicles as phytosomes. The particle size of formulated formulations varied from 122.1 nm to 998.46 nm. Prepared formulations' zeta potential ranged from -21.8 to -30.7mV. In the freeze-thaw stability analysis of phytosomal formulation F8, 10% dextrose was found to be a suitable cryoprotectant, as was the least particle Z average and PDI (569.2 and 0.79). In SEM research, vesicle size was between 463 nm and 794 nm. DSC and XRD experiments have confirmed phytosome formation. Cassia fistula extract phytosomes is a novel delivery mechanism developed and characterised in this research. The optimised F1 formulation shows promising results in terms of particle size, zeta potential, stability, SEM, DSC and XRD analysis.
