Proteomics-based analysis of lung adenocarcinoma associated protein.

- Abstract
Objective To detect the protein markers related to lung adenocarcinoma for early diagnosis, monitoring and treatment of lung adenocarcinoma. Methods Soluble proteins extracted from lung adenocarcinoma and adjacent normal tissues were subjected to two-dimensional gel electrophoresis. Then the proteins over-expressed in lung adenocarcinoma were screened and idenified by matrix-assisted laser desorption/ionization time-of-flight mass spectrum (MALDI-TOF MS) and bioinformatic techniques. The expression of grp94, one of the over-expressed proteins, was confirmed by immunohistochemistry technique. Results Six proteins including grp94 were identified by proteomic technology. These proteins were significanlly over-expressed in lung adenocarcinoma issues compared with those in adjacent normal issues. The high-expresiion of protein grp94 was confirmed by immunohistochemistry technique. The functions of the identified proteins involved intracellular protein synthesis or assembly, removal of superoxide radicals in the body, and apoptosis, which are closely related to tumor biological characteristics. Conclusion Proteomic technology can effectively screen the protein markers of lung adenocarcinoma. These proteins include Cu, Zn superoxide dismutase; heat shock protein 27; chain B, triosephosphate isomerase; annexin I; 94 ku glucose-regulated protein; and heat shock protein 90-beta. They may be new targets for molecular diagnosis and treatment of lung adenocarcinoma.
