- Abstract
To study the anti-tumor activity on HeLa cells in vitro and the anti-tumor mechanism of Sarcodonin G, a diterpenoid isolated from the dichloromethane extract of Sarcodon scabrosus karst, the effect of Sarcodonin G on HeLa cancer cell lines was determined by MTT assay in vitro. The influence of Sarcodonin G on the HeLa cell apoptosis was studied with fluorescent staining technique. The morphological changes of HeLa cells were observed by transmission electron microscope. Sarcodonin G could obviously inhibit the proliferation of cultured HeLa cells. The IC50 was 7.19 µmol/L, and it existed good relationship with the dosage;the apoptosis of HeLa cells induced by Sarcodonin G showed a marked difference in untreated HeLa cells ( P < 0.01 ). The morphological changes of HeLa cells incubated with this drug showed chromatin condensation, marginal convergency and nuclear fragmentation. Sarcodonin G can inhibit the growth of HeLa cells in vitro. It is inferred that the anti-tumor activity of Sarcodonin G was achieved by inducing HeLa cell apoptosis.
