- Abstract
This article presents information related to preparation of puromycin-resistant primary embryonic fibroblast feeder layers in transgenic mice. Transgenesis in the mouse has progressed from single gene methods to multiple targeting strategies, driving the search for additional drug selection markers. The puromycin resistance gene, puromycin N-acetyltransferase, which has been used successfully in mammalian cell lines, should prove valuable to mouse transgenics. The use of puromycin in cell lines as a selection agent is advantageous, since it achieves cell death in 2 days compared with the 7-10 days required for the commonly used neomycin and hygromycin.
