- Abstract
To study the biodegrading mechanism and application of cembratriene-4,6-diol (CBT) degrading bacteria in reconstituted tobacco,a high efficient CBT degrading strain H3-1 was isolated from tobacco leaf by spread plate method. As the sole carbon and energy source,85. 85% CBT can be degraded with this bacterium strain in 30 h, and the main degradation product is farnesal. In addition, when the maltose (2 g/L) was added to the medium,the degradation product contained solanone. The strain H3-1 was identified preliminarily as Novosphingobium sp through the analysis of classical morphology and ITS sequence of phylogenetic tree. Adding strain H3-1 (5%) in reconstituted tobacco concentrate solution, The results showed that the concentration of benzyl alcohol, 2-acetylpyrrole, 3-hydroxy-2-methyl-4H-py-ran-4-one, benzyl alcohol, 2-ethyl-3 -methyl-maleimide, methyl-cyclopentanone, solanone, dihydroactinidi-olide, 4-hydroxy-β-damascone, 9-hydroxy-4, 7- giant bean three ketene, 4-(3-hydroxybutyl)-3,5, 5-trime-thyl 2-cyclohexene-l-ketone, phytol and palmitic acid were increased after 24 h, and smoking results showed that the concentration of aroma and aroma quantity were improved. In conclusion, Novosphingobi-um sp H3-1 can improve the smoking quality of reconstituted tobacco through degrading CBT into farnesal and solanone.
