- Abstract
The β-glucan of Ganoderma lucidum was sulfated under different temperatures(40, 50, 60, 70, 80, and 90 °C)by the chlorosulfonic acid method to produce sulfated derivatives SGLP1~SGLP6. As the reaction temperature increased, the degree of substitution(DS)of SGLP1~SGLP6 initially increased and peaked at 1.72 under 60°C, and then gradually decreased. The solubility of sulfated derivatives ranged between 0. 901 and 1.102 g/100 mL, which was about 50 times of that of the non-sulfatedβ-glucan. SGLP1~SGLP6 showed anti-inflammatory activity by inhibiting NO production in lipopolysaccharide(LPS)-induced RAW264.7 cells. The inhibitory activity on NO production was concentration dependent and positively associated with the DS of the sulfated derivatives. SGLP1~SGLP6 also inhibited the phagocytosis of LPSinduced RAW264.7 cells in a DS-dependent manner. Among sulfated derivatives with similar DS, derivatives with a greater weight average molecular weight displayed higher inhibitory activities on NO release and phagocytosis in LPS-induced RAW264.7 cells, suggesting that the in vitro anti-inflammatory activity of the sulfated polysaccharides was associated with DS and weight average molecular weight.
