- Abstract
Objective: To investigate the effects on Heterocephalus glaber alveolar epithelial type II cells (AEC II) related factors mRNA expression at different concentration of aflatoxin (AF) and explore the molecular mechanism of Heterocephalus glaber against lung cancer. Methods: The lung tissue of Heterocephalus glaber was digested with enzymes and purified by immune adherence for primary culture. After being cultured for 40 h, the primary cultured cells were randomly divided into 6 groups: AF were added to the medium at final concentrations of 0.25, 0.5, 1.0, 2.0 and 4.0 mg/L as AF treated groups, DMSO (0.4 mL/L) was added to the mediumof solvent control group. The cells were incubated with AF or DMSO for 48 h, and harvested. The expression levels of SP-C, TNF-α, IL-1, IL-6, IL-8 and IL-12 in AEC II cells were detected with real-time quantitative PCR. Results: AEC II cells of Heterocephalus glaber were harvested with a purity of above 70 %and a viability of above 90 %, which meeted the requirement in vitro experiments. Quantitative PCR results showed AF made the expression levels of SP-C in Heterocephalus glaber AEC II cells significantly decreased. In the low concentrations of AF groups, the expression levels of TNF-α, IL-1, IL-6, IL-8 and IL-12 in Heterocephalus glaber AEC II cells showed no statistically significant difference as control. Conclusions: In the case of cell damage caused by AF, Heterocephalus glaber AEC II cells remained relatively low levels of inflammation factor expression. This might be one of the resistance mechanismagainst lung cancer.
