- Abstract
Objective To assess DAZL (Deleted in AZoospermia-Like) gene expression in spermatozoa of infertile men. On the human, DAZL has reported as a candidate autosomal gene for male infertility. In this study we investigated whether that gene express or not in infertile men Methods The expression of DAZL mRNA in the spermatozoa of infertile men by reverse transcription-polymerase chain reaction (RT-PCR). Also we performed to analyze Y chromosome microdeletion about genomic DNA. Spermatozoa were obtained from two infertile men who visited our hospital. Five blood samples were obtained from another infertile patients who were azoospermic men. The informed consent was obtained from all patients. After cDNA synthesize from spermatozoa, samples were examined using RT-PCR Results We examined three genetic tests for infertile men. 1. We have not detected DAZL mRNA in mature spermatozoa in this time. 2. Y chromosome microdeletion was not detected from five azoospermic men. 3. On the single spermatozoa also we have not detected deletion Conclusion Infertile men have possibility genetic disorder such a DAZL gene on autosomal chromosome. That is important in transmission of infertile gene from Mother to son. To analyze for DAZL mRNA is one of useful tool for screening of parents/ son families to identify mutation transmission.
