- Abstract
The chloroplast SSR (cpSSR) molecular marker was used to analyze the genetic diversity of 64 yam germplasm resources introduced from home and abroad, and to construct a DNA fingerprinting map, which aims to provide a basis for the parent selection of yam varieties, the innovation of yam germplasm, and the variety improvement. Show: From 21 pairs of cpSSR primers, 10 pairs of primers with stable amplification and high polymorphism were selected, 69 bands were amplified from 64 yam germplasm materials, 59 polymorphic bands, polymorphism The ratio is as high as 85.51%, the average number of alleles observed (Na) is 2, the average number of effective alleles (Ne) is 1.5693 (1.2311~1.9160), the average Shannon information index is 0.5606 (0.3290~0.6711), and the average Nei's gene is diverse The sex index (He) is 0.3781 (0.1848~0.4781), indicating that 64 yam germplasm resources have rich genetic diversity. The cluster analysis results show that the genetic similarity coefficient varies from 0.55 to 0.93. When the similarity coefficient is 0.63, 64 yam germplasm resources can be divided into 4 categories. The results show that the classification of yam germplasm resources based on cpSSR molecular markers has good distinguishing ability between species, but the cpSSR classification shows a large geographic distribution correlation., And it is not closely related to morphological characteristics. The principle of choosing fewer primers to distinguish more varieties is the principle of using 2 pairs of primers to successfully construct the DNA fingerprint of the tested germplasm, which is the choice for the identification of yam germplasm resources and the selection of parents for breed selection Provide theoretical basis.
