- Abstract
Object: to study the anti-oxidative effects of taxanes, we used suitable concentration of hydrogen peroxide to establish the oxidative sk-n-sh cell model and added four taxanes compounds (Taxine B, 7-Deacetyl-taxine B, 5-Cinnamoyloxy-taxin B and 7, 10-Diacetyl-2'-deoxyl-taxine A) to intervene the oxidative stress 2 hrs before hydrogen peroxide application. Their neuroprotective effects were evaluated using 3-(4,5)-dimethylthiahiazo (-z-yl)-3, 5-di-pbenytetrazoliumromide (MTT) assay, their effects of alleviating the nuclear morphological changes were detected after Hoechst 33258 staining and the activity of decreasing intracellular reactive oxygen species (ROS) was surveyed with flow cytometry. We discovered that 7-Deacetyl-taxine B and 5-Cinnamoyloxy-taxin B significantly improved cell viability, decreased obviously the number of cells with pycnosised neuclei and reduced intracellular ROS remarkably comparing with oxidative stress model. So we got the conclusion that 7-Deacetyl-taxine B, 5-Cinnamoyloxy-taxin B can reduce intracellular ROS, protect ceils neucleis and improve viability of oxidative model cells.
