- Abstract
The mycelia of F. filiformis Dan3 were treated with different concentrations of α-ketoglutaric acid. The results showed that the mycelia growth was accelerated by low concentrations of α-ketoglutaric acid (1 ~ 5 mmol/L) and inhibited by high concentrations (10 ~ 20 mmol/L). Further study showed that the expression levels of three genes (HCS, HIDHand SDH) in the lysine synthesis pathway of F. filiformis increased when the concentration of α-ketoglutaric acid was low, but decreased when the concentration of α-ketoglutaric acid was high. The expression levels of four other genes (HAH, AAR, AAT2and SDR) decreased after addingα-ketoglutaric acid. The lysine content in F. filiformisshowed the same trend as increased at low concentration of α-ketoglutaric acid and reduced at high concentration. 2.5 mmol/L was the best concentration to induce lysine synthesis.
