Detection and elimination of grapevine leafroll-associated viruses (GLRaVs) from Vitis vinifera cultivars of Pakistan by meristem tip culture.

- Abstract
The present study aimed to detect and eliminate Grapevine leafroll-associated viruses (GLRaVs) in commercially available Vitis varieties in order to produce appropriate clones for certification. A total of nine commercially available grapevine (Vitis vinifera L.) cultivars grown in the Lahore vineyards of Pakistan were selected. Real-time RT-PCR (TaqMan) assay was used before and after the regeneration of vine plants for the detection of GLRaVs. All nine cultivars were infected with two or more GLRaVs before apical meristem tip culture therapy. Apical meristem tip culture therapy was used as a mean to eliminate the GLRaVs from the optimum size (0.5 mm) of meristem tips. Out of 270 isolated meristems (30 each of nine cultivars), a total of 207 plants were regenerated with a 76.7% regeneration rate. The A7 cultivar demonstrated the highest regeneration rate (86.7%), while the A3 cultivar showed the lowest regeneration rate (66.7%). Screening after apical meristem tip culture therapy showed that the A1, A4, A5 & A8 cultivars demonstrated 100.0% elimination rate, while the A2, A3, A6, A7 & A9 cultivars demonstrated > 95.0% elimination rate, resulting in 97.5% overall elimination rate. The study concludes that apical meristem tip culture is an effective method for the elimination of GLRAVs infection from grapevine plants.
