- Abstract
Understanding the mechanisms of primary and secondary resistance to therapeutics that target the human epidermal growth factor family of membrane receptors (HER) for tyrosine kinase makes it possible to identify the predictive factors that determine therapeutic effectiveness or failure. The factors identified, whether for HER-1 or HER-2, fall into broad categories based on modifications to the target itself (mutated forms of the extracellular or intracellular part of the receptor), changes in the activity of receptor components, control of receptor signaling pathways (PTEN and K-RAS mutations) and, finally, the presence of related signaling pathways (IGF-1R for targeting HER-2).
