- Abstract
The purpose of the experiment was to investigate the effect of Qinghai Qaidam AC on oxidative stress injury of H9c2 rat cardiomyocytes. The cardiomyocytes of H9c2 rats were selected as the research objects, and normal oxygen group, normal oxygen +AC group, hypoxia group and hypoxia +AC group were set up. The changes of CAT, GSH-Px, SOD, MDA, HO-1, TNF-α, IL-6 and IL-10, Na+-K+ -ATPase and Ca2+ -ATPase were detected by ELISA. The results showed that compared with normal-oxygen group, the content of MDA, TNF-α and IL-6 in normal-oxygen +AC group were significantly decreased (P<0.05), and the activitiy of CAT, HO-1, Na+-K+ -ATPase and Ca2+ -ATPase were significantly increased (P<0.05). GSH-Px, SOD activity and IL-10 content were extremely increased (P<0.01). In hypoxia group, the activity of GSH-Px, HO-1 and Na+-K+ -ATPase were significantly decreased (P<0.05), the activity of CAT, SOD and Ca2+ -ATPase and the content of IL-10 were extremely decreased (P<0.01), and the content of MDA, TNF-α and IL-6 were extremely increased (P<0.01). Compared with hypoxia group, the content of MDA, TNF-α and IL-6 in hypoxia +AC group were extremely decreased (P<0.01), while the activity of CAT, GSH-Px, SOD, HO-1, Na+- K+ -ATPase, Ca2+ -ATPase and the content of IL-10 were extremely increased (P<0.01). The study indicates that AC can alleviate hypoxic injury of H9c2 rat cardiomyocytes by alleviating oxidative stress injury, enhancing energy metabolism of cardiomyocytes under hypoxia, inhibiting hypoxicinduced inflammation, and thus alleviating hypoxic injury of H9c2 rat cardiomyocytes.
