- Abstract
Brain cancer presents a complex and challenging landscape for treatment, characterized by limited efficacy of traditional therapies, the formidable barrier posed by the blood-brain barrier (BBB), tumor heterogeneity and the emergence of drug resistance mechanisms. In this review, we examine recent advancements and future perspectives in brain cancer drug discovery and targeted delivery. We explore innovative strategies such as nanoparticle-based drug delivery systems, intranasal delivery for BBB bypass, disruption techniques to enhance drug penetration and ligand-conjugated nanoparticles for targeted therapy. Additionally, we discuss personalized medicine approaches, integration of artificial intelligence in drug discovery, advancements in non-invasive imaging techniques, and the potential of gene editing technologies. Despite these advancements, challenges persist, including drug resistance, safety concerns of targeted therapies, the need for collaborative efforts and ethical considerations. We highlight the importance of interdisciplinary collaboration, regulatory adherence, and equitable access to care in navigating these challenges. By addressing these complexities and leveraging emerging opportunities, we aim to contribute to the development of more effective and personalized therapies for brain cancer, ultimately improving outcomes and quality of life for patients.
