- Abstract
Stevia glycoside is a new natural food sweetener extracted from stevia leaves and composed of a variety of diterpenoid glycoside monomers, among which rebaudioside A, rebaudioside C, steviolbioside and stevioside are well known to consumers. In this paper, stevia crystal mother liquor was used as raw material, and the separation of stevia glycoside monomer was realized by medium--pressure preparative chromatography technology. The silica gel normal phase column was used by medium--pressure preparative chromatography technology, and methanol: dichloromethane was used as eluent, which three major raw materials with RUB, DA, RC and STV as main components were obtained initially. The separation conditions of different monomers were studied by using acetonitrile and water as eluent through C18 reversed phase column. By the way, the purities of RUB, DA, RC and STV were all more than 80%, and the purity of each monomer was more than 95% when combined with recrystallization. Then the sweetness characteristics of the isolated monomers were studied to provide theoretical support for the development and application of stevia in China
