IL-13 RECEPTOR IN PSORIATIC KERATINOCYTES: OVER-EXPRESSION OF THE mRNA AND UNDER-EXPRESSION OF THE PROTEIN.

- Abstract
Although several cytokines and their receptors have been involved in the development of psoriasis, the etiology is still unknown. In this work we looked for genes possibly involved in the disease by the RT-PCR differential display technique in lesional and nonlesional skin from psoriatic patients. We found the mRNA of IL-13Ralpha1 expressed differentially in psoriatic biopsies. By RT-PCR, we confirmed an over-expression of IL-13Ralpha1 and IL-4Ralpha mRNAs in lesional skin psoriatic, when compared to skin from healthy subjects (p<0.01). The non-lesional skin of psoriatic patients presented also an over-expression of these mRNAs in 50% of the samples. IL-13 and IL-4 were not detected either as mRNA or as the proteins in any of the biopsies from psoriatic patients. Antibody to IL-13Ralpha1 detected the receptor in the epidermal keratinocytes of psoriatic patients and of healthy subjects. However, the positive antibody reaction was stronger in skin tissue from healthy subjects than in psoriatic lesional skin tissue (p<0.01), although the mRNA was over-expressed. Since the IL-13 is a pleiotropic immunoregulatory cytokine with a variety of effects on different cell types, we suggest, based on our results, that the IL-13R possibly plays an important function in the early inflammatory process of psoriasis, but its function is lost in the psoriatic keratinocytes.
