- Abstract
Die Wirksamkeit und Verträglichkeit eines Medikaments kann verbessert werden, indem dieses gezielt an den gewünschten Wirkungsort gebracht wird. Bisphosphonate sind Prototypen derartiger Medikamente; allerdings ist ihre Affinität zum Knochen eher zu stark und lässt sich nicht modulieren, ohne die Aktivität auf das Enzym Farnesylpyrophosphat ‐ Synthase (FPPS) zu verlieren. Eine Konsequenz der hohen Knochenaffinität ist außerdem eine niedrige und stark variable orale Bioverfügbarkeit. Wir beschreiben hier Inhibitoren von FPPS mit einem Monophosphonat als Knochenanker, der eine moderate Affinität zum Knochen verleiht, die überdies noch moduliert werden kann. Über einen NMR ‐ basierten Knochenbindungstest wurde ein Zusammenhang von Struktur und Knochenaffinität etabliert. Das Konzept, Wirkstoffe durch Anhängen einer Monophosphonatgruppe und ohne Verlust der oralen Bioverfügbarkeit gezielt zum Knochen zu bringen, kann weitreichende Anwendungsmöglichkeiten auch für andere Wirkstoffe haben. Auf die Knochen: Medikamente gegen Knochenkrankheiten werden besser, wenn sie gezielt an Knochen binden. Zu diesem Zweck können Knochenanker an den gewünschten Wirkstoff angebracht werden, die dessen Wirksamkeit und Verträglichkeit erhöhen, während Zellgängigkeit und orale Bioverfügbarkeit beibehalten werden. Dies wird am Beispiel von allosterischen Inhibitoren der Farnesylpyrophosphat ‐ Synthase (FPPS) gezeigt.
