- Abstract
A analysis method of UPLC-MS/MS was developed to measure the contents of 5 alkaloids in the seasonings, including morphine, codeine,thebaine, papaverine and narcotine. The samples were extracted with acetonitrile containing formic acid 0.2%, and then purified by the method of QuEChERS. Subsequently samples was degreased using the ethyl acetate and hexane(2:8, V/V), and then separated by the column of Waters UPLC HSS T3(1.7 μm, 2.1 mm×100 mm). Through gradient elution using a mobile phase consisting of 10 mmol/L ammonium formate(with 0.1% formic acid)-acetonitrile. The contents of five alkaloids were determined according to the electrospray positive ionization(ESI+) mode under multiple reaction monitoring(MRM) and quantified by external standard method. The results showed that the target compounds about 5 alkaloids had obvious peaks within 5 min,and the separation effect was good. The relative standard deviation(RSD) of the precision test was 0.43%-1.85%, the average recoveries about the five alkaloids ranged from 65.7%-108.2% with relative standard deviations(RSDs) from 1.24% to 8.71%. The limits of detection(LOD) and limit of quantitative(LOQ) for morphine, codeine were 3.0 μg/kg, 10.0 μg/kg, respectively. The LOD and LOQ for thebaine, paverine, and narcotine were 0.6 μg/kg,2.0 μg/kg, respectively. This method was simple, fast, accurate, and was suitable for the qualitative and quantitative analysis of five alkaloids of poppy husk in seasonings.
