- Abstract
objective: To investigate the protective effect and possible mechanisms of ginkgolide B on astrocytes induced by oxidative damage. Methods: Astrocytes were subcultured and divided into negative control group (cultured with normal medium), oxidative injury group (100 μmol·L-1 H2O2 treated for 12 h), ginkgolide B low dose group (1× 10-6 mol·L-1 ginkgolide B treated for 24 h, then 100 μmol·L-12O2 treated for 12 h), ginkgolide B high dose group (1× 10-4 mol·L-1 ginkgolide B treated for 24 h, then 100 μmol·L-12O2 treated for 12 h), cell viability was assayed by MTT colorimetric, reactive oxygen species (ROS) levels were detected by flow cytometry, content of superoxide dismutase (SOD) glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) in supernatants were detected by spectrophotometer.results: Ginkgolide B could inhibit oxidative damage induced cell activity decline, decreased the generation of ROS in astrocytes, promote the rise of levels of SOD, GSH-Px and decline of MDA.conclusion: Ginkgolide B play its strong antioxidant effect by increasing the intracellular content of SOD, GSH-Px levels and decreasing MDA levels, which provide reliable experimental basis for treatment of cataracts.
