桃NAC家族基因生物信息学分析及其响应低温胁迫的表达特征.

- Abstract
NAC (NAM-ATAF1/2-CUC2) are transcription factors with multiple biological functions, they play an important role in plant response to abiotic stress, hormone signal response and organ formation. In order to understand peach NAC protein (PpNAC) and its response during low temperature stress, the members, structure, function and expression of peach NAC family gene under low temperature stress were analyzed by bioinformatics and qRT-PCR. The results showed that there were 119 members in NAC family of peach, named as PpNAC001- PpNAC119. The amino acid length of PpNACs ranged from 68 to 862, with an average of 352 aa. Molecular weight ranged from 7.2 to 96.1 ku, and isoelectric point ranged from 4.3 to 9.5. Except for PpNAC119, the rest genes were located on eight chromosomes of peach. The analysis of gene structure and conserved motif showed that gene structure of the members in the same subgroup was similar, the number and length of exons were basically the same, and types and sequence of motifs contained in the members in the same subgroup were similar. The differences among different subgroups might be related to their functional specificity. The cis-acting elements in 2 000 bp sequence of PpNACs promoter was analyzed. The results showed that abscisic acid responsive, light responsive, MeJA-responsive, low-temperature responsive and auxin responsive elements accounted for a large proportion of all the selected elements. qRT-PCR analysis showed that the expression level of some genes was not significantly up-regulated compared with the control under 4 ℃ low temperature stress for 2 and 6 h. The expression levels of 16 NAC genes were significantly up-regulated after 12 h of low temperature stress.
