- Abstract
The domestic pigeon is a kind of monomorphic bird. There is almost no difference between male and female pigeons on their appearances. Therefore, the sex identification in pigeons is difficult. The nestling pigeon must be put on a foot ting at around 5 ~ 7 days old. It is necessary to develop a sex identification kit for differentiating gender in a week. The male has a pair of Z chromosomes, and the female have one Z and one W chromosome. Since the two sex chromosomes consist of its DNA sequences, and RPA was used in this study. The fluorescent primers W2-F and W3-R were designed based on W chromosome, and were been used to detect sex identification. The results of RPA were performed by the NARD kit. Two bands are visible in the female; only one band in the male. This method has high accuracy in sex identification. The traditional detection process takes time and requires the use machine. In the future, this method could be developed into a kit of rapid pigeon sex identification on farm.
