Analysis of ultrasound-assisted extraction of polyphenols from Moringa oleifera leaves and its antioxidant activity.

- Abstract
Ultrasonic-assisted extraction was used to extract polyphenols from Moringa oleifera leaves with ethanol as solvent. Single-factor experiment combined with Box-Benhnken response surface method were used to optimize the extraction conditions. According to the optimum extraction conditions, the content of polyphenol extracted from Moringa oleifera leaves of different geographic origins were determined. The antioxidant activity of polyphenol from different Moringa oleifera leaves was determined by ABTS and FRAP methods in vitro, and the cellular antioxidant activity was measured on PC-12 cells by MTT method. The experimental results showed that the optimal ultrasonic-assisted extraction conditions were ethanol concentration 80%, solid-liquid ratio 1:30 (g/mL), temperature 51°C, time 52 min. Total polyphenol content extracted from Moringa oleifera leaves of different geographic origins: Europe Moringa oleifera > India Moringa oleifera > Africa Moringa oleifera > Yunnan Moringa oleifera, and total polyphenol content extracted from Europe Moringa oleifera leaves was the highest, reached 29.11 mg GAE/gDW. The antioxidant activity analysis in vitro of polyphenol extracted from Moringa oleifera leaves of four geographic origins showed that polyphenol from leaves of Europe Moringa oleifera showed strong antioxidant activity. In this study, an extraction method of Moringa oleifera leaves polyphenols was established and it was found that Moringa oleifera leaves polyphenols could be used as a natural antioxidant with high application value and development prospect.
