- Abstract
This article describes two modifications of a whole-embryo method of preparation of barley seed for examination for loose smut, <em>Ustilago nuda </em>. Extraction without dehulling has the advantage that the whole process can be carried out in two days with a total working time of about 1-1 ½ hours, but it has the disadvantage that a good supply of hot water must be readily available. The second method, extraction after dehulling with acid, uses very little hot water and requires no special apparatus, but the working time is slightly greater.
