Isolation of an Escherichia coli strain mutant unable to form biofilm on polystyrene and to adhere to human pneumocyte cells: involvement of tryptophanase.

- Abstract
Escherichia coli adherence to biotic and abiotic surfaces constitutes the first step of infection by promoting colonization and biofilm formation. The aim of this study was to gain a better understanding of the relationship between E. coli adherence to different biotic surfaces and biofilm formation on abiotic surfaces. We isolated mutants defective in A549 pneumocyte cells adherence, fibronectin adherence, and biofilm formation by random transposition mutagenesis and sequential passages over A549 cell monolayers. Among the 97 mutants tested, 80 were decreased in biofilm formation, 8 were decreased in A549 cells adherence, 7 were decreased in their adherence to fibronectin, and 17 had no perturbations in either of the three phenotypes. We observed a correlation between adherence to fibronectin or A549 cells and biofilm formation, indicating that biotic adhesive factors are involved in biofilm formation by E. coli. Molecular analysis of the mutants revealed that a transposon insertion in the tnaA gene encoding for tryptophanase was associated with a decrease in both A549 cells adherence and biofilm formation by E. coli. The complementation of the tnaA mutant with plasmid-located wild-type tnaA restored the tryptophanase activity, epithelial cells adherence, and biofilm formation on polystyrene. The possible mechanism of tryptophanase involvement in E. coli adherence and biofilm formation is discussed.Key words: Escherichia coli, biofilm, adherence, A549 cells, fibronectin, tryptophanase.L'adhérence de Escherichia coli à des surfaces biotiques et abiotiques constitue la première étape du processus infectieux en favorisant une colonisation bactérienne et à la formation de biofilm. L'objectif de cette étude a été d'en savoir plus sur la relation entre l'adhérence à différentes surfaces biotiques et la formation de biofilm sur une surface abiotique chez E. coli. Nous avons isolé des mutants défectueux pour leur adhérence aux cellules pneumocytaires A549, pour leur adhérence à la fibronectine et pour leur capacité de formation de biofilm sur du polystyrène par mutagénèse par transposition aléatoire et par passage répété sur des monocouches de cellules A549. Parmi les 97 mutants testés, 80 avaient une diminution dans leur formation de biofilm, 8 avaient une diminution dans leur adhérence aux cellules A549, 7 étaient diminués pour leur adhérence à la fibronectine et 17 n'étaient perturbés dans aucun des trois phénotypes testés. Nous avons observé une corrélation entre l'adhérence à la fibronectine ou aux cellules A549 et la formation de biofilm, ce qui indique que les facteurs bactériens d'adhérence aux surfaces biotiques sont également impliqués dans la formation de biofilm chez E. coli. L'analyse moléculaire des mutants a montré qu'une insertion de transposon dans le gène tnaA codant pour la tryptophanase était associée à une diminution conjointe de l'adhérence aux cellules A549 et de la formation de biofilm chez E. coli. La complémentation du mutant tnaA avec un gène tnaA plasmidique sauvage a restauré l'activité tryptophanase, l'adhérence aux cellules épithéliales et la formation de biofilm sur polystyrène. Le mécanisme possible de l'intervention de la tryptophanase dans l'adhérence et la formation de biofilm chez E. coli est discuté.Mots clés : Escherichia coli, biofilm, adhérence, cellules A549, fibronectine, tryptophanase.
