- Abstract
Thermophilic bacteria are refractory and widely spread, relatively in elevated temperature habitats such as in hot spring. Isolation of novel thermophiles for biotechnological and industrial usage has come into prominence in recent years because of their ability to possess unique enzymes under extreme circumstances having high thermal activity and stability at high temperatures. The aims of this study are the isolation of thermophilic and screening of the extracellular enzymes from thermophilic bacteria using conventional methods (morphological, physiological and biochemical tests), and screening for the production of extracellular enzymes. The thermophiles bacteria were isolated from water samples and identified from Sungai Klah Hot Spring. Fifteen samples were then selected for further study based on the difference in morphological characteristics. The isolates were found to be Gram-positive, rod-shaped bacilli which able to grow at temperatures ranging from 30°C to 60°C and pH 6-9. A total of 15 isolates was able to grow on between 1-12% NaCl (w/v), and 11 out of 15 showed less growth at 14 to 16%, and no growth was observed at 18% NaCl (w/v). The results suggested that the isolates belong to the moderate halophiles group and facultative thermophiles. Furthermore, screening results of the isolates showed that three isolates (SungB2, SungB3 and SungC2) have the strongest enzyme activity for amylase, lipase, pectinase, cellulose, protease and xylanase which later selected for the enzyme production. Moreover, enzymatic products of these isolate worth further attention due to their potential applications in the field of biotechnology.
