Enrichment and characterization of MTBE-degrading cultures under iron and sulfate reducing conditions.

- Abstract
The aim of this study was to enrich cultures capable of anaerobic methyl tert-butyl ether (MTBE) biodegradation and to determine their compositions using biomolecular tools. MTBE biodegradation as the sole carbon source was observed under Fe(III) and SO42- reducing conditions and with both electron acceptors present together. The estimated MTBE biodegradation rates ranged from 3.80 to 9.20 mg·L-1·d-1 when Fe(III) was the sole electron acceptor, from 1.46 to 1.70 mg·L-1·d-1 when Fe(III) and SO42- were present together, and from 1.13 to 1.71 mg·L-1·d-1 when SO42- was the sole electron acceptor. Five to eight members were identified in the three consortia, and their characteristics were congruent with the electron acceptor conditions. A clone 99% similar to a recently described MTBE degrader, Ochrobactrum cytisi, was detected in both cultures containing Fe(III). Other 16S rRNA gene sequences detected were highly similar at the species or genus level to additional known MTBE degraders, including Pseudomonas spp. (cometabolic), Sphinogomonas, Achromobacter, and Rhodococcus. Results suggest that the buildup of intermediates and (or) the presence of sulfides had an inhibitory effect on biodegradation. Anaerobic MTBE biodegradation remains poorly understood, and degrading strains still have not been identified to date. This work represents the first detailed 16S rRNA gene profiling of highly enriched iron- and sulfate-reducing MTBE-degrading consortia. These results may provide useful biomarkers to support current efforts for confirming anaerobic MTBE biodegradation in the field. La présente étude visait à enrichir les cultures pouvant dégrader l’éther tert-butylique méthylique (MTBE) en anaérobie et à déterminer leur composition au moyen d’outils biomoléculaires. La biodégradation du MTBE comme source unique de carbone a été observée sous des conditions réductrices pour le Fe(III) et le SO42- et quand ces deux accepteurs d’électrons sont présents conjointement. Les taux estimés de biodégradation du MTBE variait entre 3,8 mg l-1d-1 et 9,2 mg l-1d-1 lorsque le Fe(III) était le seul accepteur d’électrons, entre 1,46 mg l-1d-1 et 1,70 mg l-1d-1 lorsque le Fe(III) et le SO42- étaient présent ensemble, et entre 1,13 mg l-1d-1 et 1,71 mg l-1d-1 lorsque le SO42- était le seul accepteur d’électrons. Cinq à huit membres ont été identifiés dans les trois consortiums et leurs caractéristiques concordaient avec les conditions d’accepteurs d’électrons. Un clone similaire à 99 % à un agent de dégradation du MTBE récemment décrit, Ochrobactrum cytisi, a été détecté dans les deux cultures contenant du Fe(III). D’autres séquences d’ARNr 16s détectées étaient très semblables au niveau des espèces ou des gènes à d’autres agents connus de dégradation du MTBE, dont Pseudomonas spp. (cométabolique), Sphinogomonas, Achromobacter et Rhodococcus. Les résultats montrent que l’accumulation d’intermédiaires et (ou) la présence de sulfures avaitent un effet inhibiteur sur la biodégradation. La biodégradation anaérobie du MTBE reste mal comprise et, à ce jour, les souches de dégradation ne sont toujours pas identifiées. Le présent travail représente le premier profilage détaillé d’ARNr 16s de consortiums fortement enrichis en fer, réducteurs de sulfates et dégradant le MTBE. Ces résultats peuvent fournir des biomarqueurs utiles pour soutenir les efforts actuels visant à confirmer la biodégradation anaérobie du MTBE sur le terrain.
