- Abstract
Parkinson's disease is a chronic, progressive neurodegenerative disorder whose exact cause is unknown as well as its cure. Drosophila melanogaster has widely been used as a model to study a number of neurodegenerative diseases. Exposure to paraquat is employed to induce oxidative stress induced neurotoxicity and sporadic Parkinson's disease. In this study, the therapeutic anti-oxidant properties of two flavonoids, fisetin and hesperidin against paraquat induced oxidative damage and neurotoxicity in Drosophila melanogaster has been examined. Significant locomotor disability was observed among adult female (24.3 ± 1.5%) and male (37.7 ± 2.4%) flies (CsBz strain) on exposure to 7 and 12 mM paraquat solution respectively for 12 hours. More than 45% of flies fed on flavonoids survived as compared to 100% mortality observed in paraquat treated flies at 24 hours. Also, the locomotor activity of paraquat exposed flies was enhanced by 80-85% when treated with fisetin and hesperidin for 24 hours. Fisetin and hesperidin ameliorated intracellular ROS levels, superoxide dismutase and catalase as compared to the levels increased by paraquat. They also reduced acetylcholine esterase activity which was increased by paraquat. The levels of reduced glutathione which were diminished by paraquat were also enhanced by fisetin and hesperidin. This study demonstrated that feeding flies with fisetin and hesperidin after exposure to paraquat showed anti-oxidant and neural protective effects leading to recovery of locomotor behaviour and extension of life span. The project confirmed the utility of Drosophila melanogaster as a model in screening putative therapeutic molecules prior to their use in mammalian models.
