Spectrum-effect relationship analysis of Wuziyanzong pill on sperm damage protection.

- Abstract
Objective To study the spectrum-effect relationship of Wuziyanzong pill on sperm damage protection. Methods The fingerprint of 12 batches of Wuziyanzong was established by UPLC method, and similarity evaluation was simultaneously performed with Similarity Evaluation System of Traditional Chinese Medicine Chromatographic Fingerprint. The common peaks were identified by comparing with the peaks of mixed chemical reference standards. Sperm suspension was prepared, and the oxidative damage model was induced by H202 system-produced reactive oxygen species (ROS). The effect of each batch of Wuziyanzong pill on the level of superoxide dismutase ( SOD) in the sperm model were investigated with vitamin C as the positive control. The spectrum-effect relationship of Wuziyanzong pill on sperm damage protection was elucidated based on the grey correlation analysis and the active ingredients were verified. Results There were 17 common peaks in the 12 batches of samples, and the similarity with the control fingerprint was more than 0. 9. A total of 12 common peaks were identified by comparison with the peaks of the mixed chemical reference standards including genipiganic acid, chlorogenic acid, ellagic acid, hypericin, isoquercetin, verbascoside, astragalin, quercetin, kaempferol, gomisin A, schisandrin A and schisandrin B. Each batch of Wuziyanzong all significantly increased the content of SOD, and showed a certain protection effect on sperm oxidative damage. Grey correlation coefficient showed that the synergistic effect of multiple components, and the contribution of hypericin, chlorogenic acid and isoquercetin to pharmacodynamic activity was 0. 85, 0. 82 and 0. 78, respectively. Different concentrations of hypericin all increased the level of SOD, and the activity of SOD in medium and high dose groups was significantly higher than that in model group ( P < 0. 001) . Conclusions The protective effect of Wuziyanzong pill on sperm damage attributed to the synergistic action of several components. The identified hyperoside, chlorogenic acid and isoquercetin were major components that contributed to antioxidant effect, and the protective effect of hyperoside on sperm oxidative damage was verified.
