- Abstract
Introducción. Las infecciones fúngicas invasivas causadas por especies de Candida, algunas de ellas resistentes a antifúngicos, representan el 88 % de las micosis en Colombia a nivel intrahospitalario y afectan principalmente a pacientes en unidades de cuidados intensivos. Los péptidos antimicrobianos han adquirido gran importancia por su gran capacidad terapéutica contra microorganismos multidrogorresistentes. Objetivo. Evaluar la actividad terapéutica in vitro e in vivo del péptido sintético PNR20 frente a especies de Candida de importancia clínica. Materiales y métodos. La actividad citotóxica del péptido PNR20 se evaluó mediante ensayos de viabilidad celular sobre células Vero. Se evaluó la capacidad del péptido para inducir cambios morfológicos e inhibir la formación de pseudohifas sobre las cepas 132251 de Candida auris y la cepa de referencia ATCC-10231 de Candida albicans, mediante microscopía de barrido y de fluorescencia. Finalmente, se analizó la carga fúngica y la histopatología de ratones BALB/c infectados con estas especies de Candida y tratados con PNR20. Resultados. La viabilidad celular in vitro no se vio afectada por el péptido PNR20. El péptido mostró un efecto antifúngico contra levaduras de C. albicans ATCC-10231 y C. auris 132251. El modelo in vivo mostró una reducción significativa de la carga fúngica en riñón, bazo e hígado de los ratones infectados y tratados con el péptido PNR20 en comparación con el grupo control. Conclusión. El uso del péptido PNR20 es una promisoria alternativa terapéutica en el tratamiento de la candidiasis diseminada.
