- Abstract
A clonagem animal pela transferência nuclear de células somáticas é uma das mais recentes biotecnologias reprodutivas utilizadas na indústria equina. Embora avanços significativos tenham sido realizados, aspectos do desenvolvimento placentário precisam ser esclarecidos para aumentar a eficácia da clonagem equina. Neste sentido, a estereologia (modelagem matemática) é uma importante ferramenta, pois possibilita avaliar a relação entre a estrutura da placenta e sua função in vivo. Objetivou-se neste estudo compreender as diferenças volumétricas das macrorregiões placentárias: corno gravídico (CG), corno não gravídico (CnG) e corpo do útero (CP) de gestações de clones equinos viáveis (CL) e clones equinos natimortos/óbitos neonatais até 12 horas após o nascimento (CLOB). Para tal, cinco placentas que geraram CL e cinco que geraram CLOB foram utilizadas. Após o delivramento, as membranas corioalantoides (MC) foram dispostas em letra "F" e fotografadas com escala de avaliação de área de superfície. Por meio do software Image J®, as imagens foram analisadas utilizando o sistema de teste de pontos incidentes em cada macrorregião. A fração de volume de cada região placentária (Vvregião) foi calculada dividindo-se a somatória dos pontos incidentes sobre a macrorregião de interesse (Ʃptarea) pela somatória de pontos incidente sobre a MC como um todo (Ʃptplac). Os dados foram submetidos à ANOVA, utilizando o teste de LSD (p ≤ 0,05). Os resultados demostraram que o volume total em cm3 das MC oriundas de CL foi menor (67,6 ± 6,6) em comparação aos animais CLOB (111,8 ± 2,2; p < 0,0001). Na análise volumétrica de cada grupo de animais nas suas respectivas macrorregiões, o CL apresentou os seguintes valores: CG (10,6 ± 1,5b), CnG (10,0 ±1,3b) e CP (47,0 ± 4,0a), p < 0,000. Já o grupo CLOB: CG (14,6 ± 1,3b), CnG (15,4 ± 0,8b) e CP (81,8 ± 2,3a), p < 0,0001. Verifica-se, portanto, que não houve diferença volumétrica entre as macrorregiões CG e CnG, mas a região de CP apresentou maior volume por cm3 em ambos os grupos de animais em contraste ao CG e CnG. Quando comparadas as macrorregiões em cada grupo de animais, CL e CLOB respectivamente, a região CG obteve os valores de 10,6 ± 1,5 e 14,6 ± 1,3 (p = 0,0875), demonstrando uma tendência de aumento no volume em cm3 no grupo de CLOB; na região CnG, CL e CLOB apresentaram respectivamente os valores de 10,0 ±1,3 e 14,4 ± 0,8 (p = 0,0139), expressando aumento de volume em cm3 no grupo CLOB. Para a região CP, CL e CLOB apresentaram respectivamente os valores de 47,0 ± 4,0 e 81,8 ± 2,3 (p = 0,0001), evidenciando um aumento no volume em cm3 também no grupo CLOB. Em suma, o grupo CLOB obteve maior volume total em cm3 da MC e especificamente nas regiões de CnG e CP quando comparado com o grupo CL. Isso pode estar relacionado a um remodelamento estrutural das macrorregiões placentárias (volume total e regional placentário) na tentativa de aumentar a eficiência de trocas maternofetais em gestações clones comprometidas e que resultaram em óbito neonatal.
